Title: Sting/Yup 50 won't start

I recently ordered a tecnigas next r for my bike,I put it on and went for 1 mile of test run,felt like losing power at WOT ,.I knew I had to upjet anyway,came home and upjet my main  to #90 from #72.I put everything back together and as I start the scoot ,It won't start .checked and replace sparkplug,siphoned gas replace with fresh one,cleaned carb and still would start...Now I'm thinking did I damage the piston in that one mile of test run?(mostly half throttle).Any advise would be appreciated.......

Finally got my sting started,thanks for Peters advice.
Basically if you drained gas out of carb from jetting/cleaning,and when you put it back together and your bike doesn't start,give it couple of kickstart (up to 20) to fill up carb with gas,holding throttle 1/4 open,just be careful not to flood engine,if you think you flooded engine wait awhile before trying again.Don't Panic like I did(lol)
